Introduction to Medicinal Chemistry

Medicinal Chemistry is a Multidisciplinary subject involving -

1. Organic Chemistry
2. Pharmacology
3. Biochemistry
4. Physiology
5. Microbiology
6. Computer Modeling
  • Medicinal Chemistry explains the design & production of compounds that can be used for prevention, treatment, or cure of human or animal diseases.

It Includes (Remember this formula - (3 I 2 D PMC) ) -

The Invention
Identification
Interpretation of their mode of action at a molecular level
Discovery
Design
Preparation of active compounds
The study of their Metabolism
Construction of structure-activity relationship

  • Beginning with AD 1800 there was a continuous activity in this area and many of the well-known Medicinal plants were analyzed and their active principle Characterized.
For example- 1. Morphine was isolated from the opium poppy
2. Quinine was isolated from cinchona species

Medicinal chemistry covers the following stages:-

1st Stage - Lead Compound - Drugs are identified and prepared from natural sources,
organic chemical reactions, or biotechnological processes.
2nd Stage - Lead Molecule - Optimization of LM to improve potency, selectivity, and to (LM) reduce toxicity
3rd Stage - Synthetic route - Optimization of synthetic route for bulk production and
modification of pharmacokinetic, and pharmaceutical properties of the active substance to render it clinically useful.

Drug Design -

Drug design based on a modification of the structure of a lead compound.
  • The efficiency of its increased recently by high -throughput screening (HTPS) system utilizing cell line culture system with enzyme-linked immune system assay (ELISA) & receptor molecules derived from gene cloning.
